AAV Capsid Engineering Workflow Details
PackGene’s Ï€-Icosa system for AAV capsid engineering offers a comprehensive AAV service for creating and screening custom AAV libraries to meet specific gene and cell therapy (GCT) needs.
- Phase I: Generate a diverse AAV capsid library through mutation and plasmid production, followed by AAV packaging, animal testing, and NGS analysis. A refined library is designed if needed.
- Phase II: Screen the updated library to identify top-performing variants with improved targeting and specificity.
- Phase III: Produce and validate the best variants through custom AAV production, large-scale animal testing, and optional histology.
This process ensures the identification of optimized AAV capsids for enhanced organ targeting and reduced off-target effects.

Excellent pre-screening library diversity and uniformity
A high-coverage AAV library with diverse and distinct capsid variants increases the chances of identifying ideal properties for AAV capsid engineering. Low coverage risks missing critical variants, limiting optimization potential.
Uniformity is equally crucial to ensure equal representation of variants, avoiding screening bias and ensuring optimal results in custom AAV production.
Extremely low mispackaging rate : <0.1%
In AAV capsid engineering, minimizing the mispackaging rate is critical for accurate screening and reliable custom AAV production. High mispackaging rates can lead to incorrect genetic material in AAV vectors, causing unintended transduction, toxicity, or reduced efficacy. Additionally, mispackaging may produce replication-competent AAV (rcAAV), posing safety risks like immune reactions in therapies for cancer or genetic disorders.
To reduce mispackaging, optimizing the vector production process is essential, including precise DNA input control, high-quality plasmid production, and refined transfection conditions. Screening for mispackaging ensures the AAV library remains representative and reliable for testing.
PackGene’s process measures the mispackaging rate by analyzing capsid protein integrity, achieving extremely low rates to match capsid genes with proteins during packaging, ensuring accurate results for AAV capsid engineering.
