Exosomes are nano-sized extracellular vesicles (EVs) ranging 30-150 nanometers. They are readily released by most cells and can be found in all body fluids including plasma, serum, saliva, cerebrospinal fluid, urine and cell culture media. Exosomes contain various biomolecules of their cell of origin, including nucleic acids, proteins, lipids, and metabolites, playing an important role in cell-to-cell communication.

 

Exosomes were first described to play a role in cellular waste disposal. Through subsequent research, more significant functions were discovered. For example, mesenchymal stem cells (MSCs)-derived exosomes have been widely investigated for their role in tissue regeneration and therapeutic effect in many diseases. Currently, exosomes have been potentially used as diagnostic and prognostic biomarkers in cancer and other diseases such as cardiovascular, tuberculosis, neurological diseases. Exosomes exhibit desirable features as an ideal drug delivery system such as low toxicity, low immunogenicity, high engineerability, high delivery efficiency and intrinsic homing ability, making it attractive for treatment of diseases such as cancer or Alzheimer’s diseases.

Therefore, characterization of exosomes can provide valuable information for detection, and disease monitoring. TRIFic™ detection assays are time-resolved immunofluorescence assays which utilize europium to measure the abundance of exosome protein markers, CD9, CD63, or CD81, in purified or unpurified samples. ExoBrite™ EV Membrane Staining Kits were designed for staining the membrane of extracellular vesicles and exosomes by providing bright, specific staining, and very low background noise for flow cytometry. The exosome markers can also be detected via western blotting using validated exosome marker antibodies, an essential tool in exosome research. Also available is our exosome-depleted FBS that provides the highest level of exosome depletion (≥ 95 %) using proprietary ultrafiltration method without compromising the cell growth performance.
